What Is Depo-Provera?
Depo-Provera is a contraceptive medication containing medroxyprogesterone acetate (a synthetic progestin hormone). It is a hormonal form of birth control that works by preventing the ovaries from releasing an egg and causing cervical mucus to thicken, helping to prevent sperm from entering the uterus. It is administered by injection in the arm or buttock every three months. Up to 25 percent of women aged 18 to 49 have used medroxyprogesterone acetate as a contraceptive, with over 74 million users worldwide.

Does Depo-Provera Cause Brain Tumors?
Multiple studies have found evidence linking the use of selected progestogens, including Depo-Provera, with an increased risk of intracranial meningioma (brain tumors). In March 2024, one study was published with data collected from 108,366 women showing an “excess risk of meningioma with use of medroxyprogesterone acetate (injectable).”
Another study in September 2024 specifically concentrated on the association between exposure to medroxyprogesterone acetate and meningioma. The findings supported previous research, confirming an increased chance of brain tumors for those using this drug, with an association that became stronger with a longer duration of use. Although the meningiomas were typically non-cancerous, they can still cause serious side effects and require surgery to treat.
While there has been no recall of Depo-Provera at this time, the first brain tumor lawsuits related to this drug were filed in October 2024. Since then, additional suits have been filed across the United States. As of October 2025, more than 1,200 of these lawsuits have been consolidated into a multidistrict litigation (MDL) suit (similar to a class action lawsuit).
